Cytogenetics of Chilean angiosperms: Advances and prospects RChHN

Cytogenetic data on Chilean angiosperms have been reported since at least eight decades ago; however, much of this information is disperse in diverse sources and is not readily available as a comprehensive document that allows having a general vision on advances and gaps in this matter. The goal of this paper is to summarize the advances and prospets on cytogenetic studies of the Chilean angiosperms based on compiled publications from 1929 to 2010. We found 78 publications supplied by four groups of Chilean researchers and some foreign specialists. Cytogenetic data have been reported for 139 Chilean angiosperm species (2.8 % of the total), which belong to 58 genera and 34 families. Detection of diploid males in a natural colony of the cleptobiotic bee Lestrimelitta sp (Hymenoptera, Apidae) Genet. Mol. Biol.

When working at quantifying the genome size of stingless bees, it was observed that males of Lestrimelitta sp possessed the same amount of nuclear DNA as the females. Thus, we used flow cytometry (FCM) and cytogenetic analysis to confirm the ploidy of these individuals. The males analyzed proved to be diploid, since, through cytometric analysis, it was demonstrated that the mean genome size of both males and females was the same (C = 0.463 pg), and, furthermore, cytogenetic analysis demonstrated that both had 2n = 28 chromosomes.

DNA content estimation of Fig and Black Mulberry using flow cytometry Rev. Bras. Frutic.

Abstract In this study, fig and black mulberry DNA contents were estimated using DAPI fluorescence stain in flow cytometry. The 2C DNA contents of the fig and black mulberry were found as 0.82 pg and 8.34 pg, respectively. The calculated 1C value of genome size of fig is 401.8 Mbp and that of black mulberry is 4086.6 Mbp. The ratio of 2C DNA content and 1C genome of the black mulberry was 10.17 times that of the fig although fig is diploid and black mulberry is decosaploid.

Genome analysis of thirteen Colombian clostridial strains by pulsed field gel electrophoresis Electron. J. Biotechnol.

Pulsed field gel electrophoresis was used for estimating the size of the genome and evaluating the presence of megaplasmids in 13 native Colombian solventogenic Clostridium strains. DNA preparation and purification were optimised for obtaining differentiated restriction fragments in electrophoresis. Genomic DNA was digested with ApaI, Eco52I, SmaI and XhoI enzymes. Estimated genome size for native strains ranged from 4.0 to 4.2 mega base pairs. Abstract Pennisetum glaucum (Pearl millet) and P. purpureum (Napier grass) are economically important members of the genus Pennisetum . The knowledge of variability in chromosome number, size and genomic content of the species could provide clues on the mechanisms responsible for decrease or increase in genomic content in the evolutionary pathway. Genome size of three Brazilian flies from the Sciaridae family Genet. Mol. Biol.

We determined the genome size of the Brazilian sciarid flies Bradysia hygida, Rhynchosciara americana and Trichosia pubescens (Diptera: Sciaridae) using absorbance measurements of Feulgen-stained nuclei belonging to these species (and chicken erythrocytes as a standard) to calculate the amount of DNA in picograms (pg) and the number of base pairs (bp), or C-value, for each of these species. The C-values were: 3 x 10(8) bp (0.31 pg) for B. hygida; 3.6 x 10(8) bp (0.37 pg) for R. americana; and 1 x 10(9) bp (1.03 pg) for T. pubescens. The sciarids investigated in this work had considerably higher C-values than the average for previously described dipteran species, including D. melanogaster.

The subgenus Ceratochloa of the genus Bromus includes a number of closely related allopolyploid forms or species that present a difficult taxonomic problem. The present work combines data concerning chromosome length, heterochromatin distribution and nuclear genome size of different 6x, 8x and 12x accessions in this subgenus. Special attention is paid to the karyotype structure and genomic constitution of duodecaploid plants recently found in South America. ABSTRACT Agapanthus (Agapanthaceae) has 10 species described. However, most taxonomists differ respect to this number because the great phenotypic plasticity of the species. The cytogenetic has been an important tool to aid the plant taxon identification, and to date, all taxa of Agapanthus L'Héritier studied cytologically, presented 2n = 30.
